@Pink Gorilla at the current moment, Joker is still whoever he was before he became Joker, so Harley Quinn would currently be Harleen Quinzel. However, the audition shall still be considered for when Harleen becomes Harley.

You can if you wish, though there is no guarantee that you will be the character. I'd advise taking a look at the https://www.rpnation.com/threads/rpnation-bbcode-guide.34813/, and then prettying up the two sheets a little more, as the better the sheet the more point you will recieve towards the character.
